Reticulocyte Counting of the Rat Blood Using Flowcytometry in Comparison with Manual Methods

Abstract

The reticulocyte counting is an important parameter in assessing of subject with anemia in laboratory animals and humans. The manual counting method ie thought to have some limitation with low precision and heavy work load. Recently fluorescent activated cytometric (FACS) analysis of reticulocytes by thiazol orange (TO) has been introduced in human hematology to improve the precision and the speed of measurement. The reticulocyte of rat blood was analyzed by manual method and FACS, and compared. A good correlation (r=0.897, slope=10.03, intercept=-3.50) between manual count and FACE determination was observed. The methodology of FACS was accurate and cost-effective, and showed a high correlation with manual techniques.
